United will find out their third opponent when the draw takes place at 11.30am on Thursday.
Karl Robinson’s side are guaranteed to play two of their three group matches at home, with the first round of games in the week commencing August 29.
United have never faced Sutton in a competitive fixture but they have history with Orient, thanks to an infamous clash between the teams on the final day of the 2005/06 season.
The U’s lost 3-2 as their relegation from the Football League was confirmed, while the East London side sealed promotion.
The teams last played each other in the 2015/16 League Two season, drawing 2-2 at Brisbane Road and Orient winning 1-0 at the Kassam Stadium.
United went out at the group stage of last season’s Trophy, beating Tottenham Under 21s after defeats to Cambridge United and Stevenage.
